2. Sub-cellular membrane proteomics
Our second task was aimed at developing different sub-cellular proteomic analyses
in order to have access to membrane proteins of the fully established AM symbiosis.
2a. First, purification and 2-DE separation protocol were designed
to gain access to the total membrane-associated proteins.

Ninety-six out of the 440 well-resolved proteins
were identified by MALDI-TOF PMF.
A high percent (83%) of successful protein identification was obtained when
using M. truncatula clustered EST database for queries.
During the purification process, the enrichment in membrane-associated proteins
was monitored on 2-DE.
The membrane location of microsomal proteins was further confirmed using
PMF identification [16]. |
